pPsiR from Agrobacterium tumefaciens
This part is the promoter region (0.36 kb upstream) of the PsiR gene of Agrobacterium tumefaciens str. C58 (Atu4743)
pPsiR is predicted to be repressed by the PsiR transcription factor (BBa_K2448006) which is inhibited in the presence of D-Psicose [1].
Usage and Biology
Our team used this pPsiR promoter together with the Agrobacterium tumefaciens PsiR transcription factor (BBa_K2448006) to build a Psicose biosensor (BBa_K2448026). The results show that pPsiR is functional in E. coli and that it behaved as predicted under and without induction.
Using the BPROM webserver [2], the predicted features of this promoter are: Threshold for promoters - 0.20 Number of predicted promoters - 1 Promoter Pos: 299 LDF- 3.13 -10 box at pos. 284 tggtgcaat Score 40 -35 box at pos. 264 ttgcca Score 61 Oligonucleotides from known TF binding sites: rpoD17: TGCAATAA at position 287 Score - 5
